  ID: TOLIP_HUMAN
DESCRIPTION: RecName: Full=Toll-interacting protein;
FUNCTION: Component of the signaling pathway of IL-1 and Toll-like receptors. Inhibits cell activation by microbial products. Recruits IRAK1 to the IL-1 receptor complex. Inhibits IRAK1 phosphorylation and kinase activity.
SUBUNIT: Oligomerizes. Binds to TLR2 and the TLR4-MD2 complex via its C-terminus. Exists as complex with IRAK1 in unstimulated cells. Upon IL-1 signaling, Tollip binds to the activated IL-1 receptor complex containing IL-1RI, IL-1RacP and the adapter protein MyD88, where it interacts with the TIR domain of IL-1RacP. MyD88 then triggers IRAK1 autophosphorylation, which in turn leads to the dissociation of IRAK1 from Tollip and IL-1RAcP. Interacts with TOM1L2.
INTERACTION: Q05048:CSTF1; NbExp=2; IntAct=EBI-74615, EBI-1789619; Q15038:DAZAP2; NbExp=3; IntAct=EBI-74615, EBI-724310; Q86Y13:DZIP3; NbExp=2; IntAct=EBI-74615, EBI-948630; P19525:EIF2AK2; NbExp=2; IntAct=EBI-74615, EBI-640775; O43187:IRAK2; NbExp=2; IntAct=EBI-74615, EBI-447733; Q86TG7:PEG10; NbExp=2; IntAct=EBI-74615, EBI-2858265; O75674:TOM1L1; NbExp=3; IntAct=EBI-74615, EBI-712991; Q96S55:WRNIP1; NbExp=2; IntAct=EBI-74615, EBI-2513471;
SUBCELLULAR LOCATION: Cytoplasm (Probable).
PTM: Phosphorylated by IRAK1 upon stimulation by IL-1 or microbial products.
SIMILARITY: Belongs to the tollip family.
SIMILARITY: Contains 1 C2 domain.
SIMILARITY: Contains 1 CUE domain.
SEQUENCE CAUTION: Sequence=BAB14283.1; Type=Erroneous initiation;

The pictures above may be empty if there is no ModBase structure for the protein. The ModBase structure frequently covers just a fragment of the protein. You may be asked to log onto ModBase the first time you click on the pictures. It is simplest after logging in to just click on the picture again to get to the specific info on that model.

-  Orthologous Genes in Other Species
  Orthologies between human, mouse, and rat are computed by taking the best BLASTP hit, and filtering out non-syntenic hits. For more distant species reciprocal-best BLASTP hits are used. Note that the absence of an ortholog in the table below may reflect incomplete annotations in the other species rather than a true absence of the orthologous gene.
