DESCRIPTION: RecName: Full=Metalloproteinase inhibitor 3; AltName: Full=Protein MIG-5; AltName: Full=Tissue inhibitor of metalloproteinases 3; Short=TIMP-3; Flags: Precursor;
FUNCTION: Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. May form part of a tissue-specific acute response to remodeling stimuli. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-9, MMP-13, MMP-14 and MMP-15.SUBUNIT: Interacts with EFEMP1.INTERACTION: P50052:AGTR2; NbExp=7; IntAct=EBI-1748085, EBI-1748067;SUBCELLULAR LOCATION: Secreted, extracellular space, extracellular matrix.DISEASE: Defects in TIMP3 are the cause of Sorsby fundus dystrophy (SFD) [MIM:136900] . SFD is a rare autosomal dominant macular disorder with an age of onset in the fourth decade. It is characterized by loss of central vision from subretinal neovascularization and atrophy of the ocular tissues. Generally, macular disciform degeneration develops in the patients eye within 6 months to 6 years.SIMILARITY: Belongs to the protease inhibitor I35 (TIMP) family.SIMILARITY: Contains 1 NTR domain.WEB RESOURCE: Name=Mutations of the TIMP3 gene; Note=Retina International's Scientific Newsletter; URL="http://www.retina-international.org/files/sci-news/timpmut.htm";WEB RESOURCE: Name=GeneReviews; URL="http://www.ncbi.nlm.nih.gov/sites/GeneTests/lab/gene/TIMP3";

mRNA Secondary Structure of 3' and 5' UTRs
The RNAfold program from the Vienna RNA Package is used to perform the secondary structure predictions and folding calculations. The estimated folding energy is in kcal/mol. The more negative the energy, the more secondary structure the RNA is likely to have.

ModBase Predicted Comparative 3D Structure on P35625
The pictures above may be empty if there is no ModBase structure for the protein. The ModBase structure frequently covers just a fragment of the protein. You may be asked to log onto ModBase the first time you click on the pictures. It is simplest after logging in to just click on the picture again to get to the specific info on that model.
Orthologous Genes in Other Species
Orthologies between human, mouse, and rat are computed by taking the best BLASTP hit, and filtering out non-syntenic hits. For more distant species reciprocal-best BLASTP hits are used. Note that the absence of an ortholog in the table below may reflect incomplete annotations in the other species rather than a true absence of the orthologous gene.
